TOM* - EICOSANOID SYSTEM AND ENDOTHELIUM:THE ROLE IN INFLAMMATION AND ATHEROSCLEROSIS

ABSTRACT


Beside eicosanoid and autocoid systems, ENDOTHELIUM represents extremely complex regulatory system, with surface more than 1000 m2.
Endothelium cells constituted major PARACRINE human organ. having a synthesis, release and/or activation of substances with procagulant, anticoagulant, fibronlytic, hemotactic, inflamatory, vasodilatory and vazoconstrictive actions. The highlights of these substances are about better understandings of autocrine, jucstacrine and paracrine controls, as well as endocrine regulation and intercellular response. The study of endothelium system and its relation with platelets, monocytes and smooth muscle cells of the vascular wall presents the main study of major research groups over the world, as well as in our country.
For the last few years it has been made a significant scientific improvement in order to highlight of HOMEOSTASIS mechanism, disorder of regulatory mechanisms in ATHEROSCLEROSIS and THROMBOSIS developing.
On the basis of research work over years we have postulated a special concept about significance of balance inbetween PGI
2 and TXA2 and their keeping fluidity and blood flow at homeostasis and preventing of ahterosclerosis and thrombosis developing.
We have been explored anti-sclerotic effects of PGI
2 besides its vasodilataory, antiaggregatory, cytoprotective and fibrinolytic effects.
PGI
2 and TXA2 misbalance have a crucial role in pathogenesis of atherosclerose as well as oxidized lipoproteins, endothelium cells, smooth muscle cells and monocytes interaction.
This system INHIBITS basic mechanism of early atherosclerosis pathogenesis such as mytogen liberation and holesterol esters accumulation. The role of eicosanoid and autocanoid systems at inflammatory process presents the theme of our investigations besides atherosclerosis and thrombosis.
